We have identified a fusion mRNA between CYCLIN D1 and TROP2 (CYCLIN D1-TROP2 chimera), that is expressed by human cell and tissues. We have shown that this chimera has a causative role in tumor transformation. This, together with the high specificity of its expression, makes the chimera an effective therapeutic target, that we have exploited to develop new highly specific and efficient anti-tumor approaches. The invention here presented concerns the identification of a fusion RNA between CYCLIN D1 and TROP2 in human cells and tissues; the methods used to reveal its presence and to quantify the levels of this fusion RNA; the use of this fusion RNA as a marker of response to therapy in human tumors; the use of this fusion RNA as a target of specific antitumor therapy; the methods used to inhibit its expression in cells and tissues; the identification and use in tumor diagnosis and antitumor therapy of other chimeric RNAs.
