The present invention concerns the identification of Trop-2 as a key component of a novel mechanism of stimulation of the growth of tumors; the identification of the central nodes of the Trop-2-dependent signals as targets of specific anti-tumor therapy; the identification of Trop-2 as predictive marker of therapeutic response to inhibitors of key molecules in these nodes, in particular of CD9, Akt and other functionally linked molecules; the use of Trop-2 to identify and select patients eligible for anti-cancer therapy directed against key molecules of Trop-2 signaling, in particular CD9, Akt and other functionally linked molecules; the methods used to inhibit the expression or to block the function of CD9 and Akt in tumors that express Trop-2 and the use of these inhibitors to selectively arrest the growth of cells expressing Trop-2.
